Gain in sensitivity and loss in temporal contrast of STDP by dopaminergic modulation at hippocampal synapses.

چکیده
Spike-timing-dependent plasticity (STDP) is considered a physiologically relevant form of Hebbian learning. However, behavioral learning often involves action of reinforcement or reward signals such as dopamine. Here, we examined how dopamine influences the quantitative rule of STDP at glutamatergic synapses of hippocampal neurons. The presence of 20 muM dopamine during paired pre- and postsynaptic spiking activity expanded the effective time window for timing-dependent long-term potentiation (t-LTP) to at least -45 ms, and allowed normally ineffective weak stimuli with fewer spike pairs to induce significant t-LTP. Meanwhile, dopamine did not affect the degree of t-LTP induced by normal strong stimuli with spike timing (ST) of +10 ms. Such dopamine-dependent enhancement in the sensitivity of t-LTP was completely blocked by the D1-like dopamine receptor antagonist SCH23390, but not by the D2-like dopamine receptor antagonist sulpiride. Surprisingly, timing-dependent long-term depression (t-LTD) at negative ST was converted into t-LTP by dopamine treatment; this conversion was also blocked by SCH23390. In addition, t-LTP in the presence of dopamine was completely blocked by the NMDA receptor antagonist 2-amino-5-phosphonovaleric acid, indicating that D1-like receptor-mediated modulation appears to act through the classical NMDA receptor-mediated signaling pathway that underlies STDP. These results provide a quantitative and mechanistic basis for a previously undescribed learning rule that depends on pre- and postsynaptic ST, as well as the global reward signal.

Effect of Sulpiride on Dopaminergic Synapse of Dorsal Hippocampus of Morphine-Treated Rats
Background: As previous studies show, several effects of morphine are induced by the dopaminergic system. Sulpiride is a dopamine D2 receptor (DAD2) antagonist widely used in clinics to treat DArelated disorders. DAD2 receptors are abundant at hippocampal cornu ammonis (CA1). Objectives: This study aimed to investigate the possible interaction of morphine and sulpiride on DA synapses in CA1...
متن کاملHuman Synapses Show a Wide Temporal Window for Spike-Timing-Dependent Plasticity
Throughout our lifetime, activity-dependent changes in neuronal connection strength enable the brain to refine neural circuits and learn based on experience. Synapses can bi-directionally alter strength and the magnitude and sign depend on the millisecond timing of presynaptic and postsynaptic action potential firing. Recent findings on laboratory animals have shown that neurons can show a vari...

Many physiological events require an accurate timing signal, usually generated by neural networks called central pattern generators (CPGs). On the other hand, properties of neurons and neural networks (e.g. time constants of neurons and weights of network connections) alter with time, resulting in gradual changes in timing of such networks. Recently, a synaptic weight adjustment mechanism has b...
متن کاملDopamine regulates intrinsic excitability thereby gating successful induction of spike timing-dependent plasticity in CA1 of the hippocampus
Long-term potentiation (LTP) and long-term depression (LTD) are generally assumed to be cellular correlates for learning and memory. Different types of LTP induction protocols differing in severity of stimulation can be distinguished in CA1 of the hippocampus. To better understand signaling mechanisms and involvement of neuromodulators such as dopamine (DA) in synaptic plasticity, less severe a...
